A novel iodixanol density gradient for purification of pancreatic islets was developed in the difficult pig model. The pig islets restored normal blood sugar levels after xeno transplantation in diabetic mice.

Efficacy of the novel iodixanol-UWS density gradient for human islet purification • Igls Aidspit 1999 • Michel van der Burg • Miracles.Media • 20241202_1
I started an archive at Archive.org November 11, 2010 with this slideshow (pdf) of my January 1999 oral presentation in Igls (Innsbruck), Austria, at the 18th Workshop of the AIDSPIT (Artificial Insulin Delivery Systems, Pancreas and Islet Transplantation) Study Group. (Now known as AIDPIT).
The presented results of this innovative islet purification method — developed over nearly 10 years (from 1989) in our Leiden islet laboratory (LUMC) — were the outcome of work I had done spring 1998 with the human islet transplantation group of Camillo Ricordi at the Diabetes Research Institute in Miami , Florida, USA.

Venue of the Seventh Annual Meeting of the Anglo–Danish–Dutch Diabetes Group*, Oxford (UK) July 5–9, 1989.
The first international meeting were our new concept was presented, of pancreatic islet isolation in the University of Wisconsin Solution (UW-Solution or UWS) a solution designed recently for preservation of a donor pancreas during cold storage prior to transplantation.
Quote :
“…A few months ago, we adopted an entirely new approach to islet isolation. Recently a new cold storage organ preservation solution was developed at the University of Wisconsin, the UW solution, allowing long term, 24 to 72 h, cold storage of the canine pancreas. Since islet isolation is largely performed in the cold, we tested this preservation solution as the isolation medium throughout the isolation procedure. This slide shows that using the UW solution, purity has increased to over 90%…”
In addition our first successful pre-clinical transplant, we had performed the last month, was reported as breaking news at the end of the talk.
Quote :
“Three weeks ago we did our first autotransplantation of isolated islets from the whole gland by retrograde infusion into the spleen. The dog is doing fine: fasting glucose amounts to 5-6 mM and postprandial glucose between 6-8 mM ; postprandial insulin increases 6-7 fold. Thank you.”

Poster creation
The poster was designed and produced May 1992 on a Macintosh computer using MacDraw II (with bar graphs from Cricket Graph) and printed landscape across a chain of several sheets of paper using the Imagewriter dotmatrix printer.
For this 2024 remastered poster below, the 1992 MacDraw II file was converted and edited using LibreOffice Vanilla, and next Pixelmator Pro was used for additional ‘repair’ (conversion issues with the many layers used for ‘tubes’ creation) and the insertion of the photo scans.

September 1989 at a symposium of two international societies in Minneapolis, USA (1) , I presented this first poster on the ‘ invention ‘ of using the UW solution ( the University of Wisconsin organ preservation solution) during pancreatic islet isolation, with the exciting discovery of near-complete purification of islets of Langerhans in the laboratory for transplantation in diabetic patients.
Here the remastered 20240804 edition of that Minneapolis 1989 poster presentation.
The UWS data had been added ad hoc in the poster – that is , it wasn’t part of the accepted abstract. The first proceedings paper on our UWS work was also submitted at this meeting , and was published April ’90 (3).
A previous edition of the remastered poster was published here two years ago (2) but that edition had a different photo print (higher magnification, and without the bar indicator) of the purified islet preparation inserted, not the original image. This has been corrected now. In addition the original line spacing is now restored.

Lancet Letter 1989 UW Solution Islet Isolation | 20220823 | Michel van der Burg | michelvanderburg•com | June 1989 preparing first submission for publication as letter in the Lancet medical journal the new concept of using the UW solution ( the University of Wisconsin organ preservation solution) for isolation and complete purification of islets of Langerhans in the laboratory for transplantation in diabetic patients. Later that year that exciting discovery was presented at meetings in Minneapolis, USA, and Barcelona, Spain. Details in yesterday’s post. Submission for publication as a letter in the Lancet had had no succes ;)

The next month at a meeting in Barcelona, Spain, this rather radical new concept was introduced (2, 3). June 1989 submission for publication as a letter in the Lancet had had no succes ;)
Poster designed and produced on my personal Macintosh ED and Imagewriter-II matrix printer, allowing full width printing over multiple pages in landscape (a low cost do-it-yourself discovery by me, at a time when posters were prepared on poster boards by graphical professionals using photography and special lettering techniques for around $400). Updates
20240804 – This remastered 20240804 edition of the Minneapolis 1989 poster , now shows the original photo of the islet prep as posted at the meeting in 1989. In addition the correct line spacing is now restored. The previous poster published online two years ago (20220822) showed a different photo print (higher magnification, and without the bar indicator) of the same islet prep, not the correct image.
