This IPITA meeting in Amsterdam was also a good opportunity to catch up with colleague Nick London (see photo) from the Leicester (UK) group discussing our Leiden and Leicester islet isolation work using the University of Wisconsin organ preservation solution (UWS). The Leicester group were one of the first groups to examine and adopt our novel method of islet isolation in UWS.
Several groups reported at this 4th IPITA symposium on the effects of using (modified) UW solution (UWS) during islet isolation : Leicester (pig, human islets), Oxford (rat, rabbit islets), Minneapolis (human islets), Paris, and Giessen (clinical human allo islet transplantation).

During the 4th IPITA I discussed in detail the rationale of our UWS work, the important components of UWS, and basal solution for islet purification (Percoll-UWS) with Yoko Mullen (in particular) and her co-worker Charles Brunicardi (see photo) of the UCLA Islet Transplant Team (LA, USA)…contributing to the development of Yoko’s Los Angeles preservation solution #1 (LAP-1) patented in 1997 (see Notes, US5679565A).

Notes

AIDPIT was known first as AIDSPIT, short for : Artificial Insulin Delivery Systems, Pancreas and Islet Transplantation.
The AIDSPIT study group of the European Association for the Study of Diabetes (EASD) was established in the early 1980s.
In 2000 the acronym AIDSPIT was replaced by ‘AIDPIT’, in order to avoid confusion with the AIDS disease.

The annual meetings are generally held in the little town of Igls, close to Innsbruck in Austria — the first years, early in February, and next, late in January. Once, in the 1990s (following a discussion ‘…why always in Igls?’, the meeting was held in Amsterdam.
From 2011, the annual workshops of AIDPIT continued as a joined meeting with the EPITA — the European Pancreas and Islet Transplant Association — part of the European Society for Organ Transplantation (ESOT).
From 2024 — following the 12th EPITA Symposium & 41st AIDPIT Workshop in 2023 — this symposium is referred to as the EPITA Symposium.

GLP-1 blood sugar regulation in islet transplants

GLP-1 (glucagon-like peptide-1) is now well-known from the anti-diabetic drugs that mimic the action of this natural hormone GLP-1, to treat type 2 diabetes and obesity. They work by stimulating insulin release, reducing glucagon secretion, slowing stomach emptying, and suppressing appetite, which leads to lower blood sugar and weight loss.

May 1992 we were the first to show direct potentiation of the insulin response from isolated islets by a (near-)physiological dose of the gut hormone GLP-1 during perifusion (in vitro) of canine islets at 7.5-10 mM glucose levels (1,2).

Jan 1994 we next showed this incretin effect of GLP-1 also after islet transplantation (in vivo, in dogs) in this poster (3) and abstract (4) presented at the 13th Workshop of the AIDSPIT (aka AIDPIT) Study Group , Igls, Austria, January 23–25, 1994.

Background of the work was posted earlier this year (5) and the work was published Nov 1994 in my PhD thesis Pancreatic islet transplantation , available online (6).

Slideshow (digitally remastered) of first talk on pancreatic islet xeno-transplantation following adult pig islet purification in our novel Iodixanol-UWS density gradient. Presented at the 17th Workshop of the AIDSPIT Study Group in Igls, Austria, Tuesday, Jan 27, 1998 (1, 2).

Isolation of porcine pancreatic islets (from the pancreas of large sows) was markedly improved by keeping the islets in the UW donor organ preservation solution (UWS) both during the isolation of islets (using the Liberase enzyme blend) and purification of the islets using a novel Iodixanol-UWS density gradient.
Transplantation of the pig islets (after 1 day storage in culture solution) restored normal blood sugar levels in diabetic mice.

I started an archive at Archive.org November 11, 2010 with this slideshow (pdf) of my January 1999 oral presentation in Igls (Innsbruck), Austria, at the 18th Workshop of the AIDSPIT (Artificial Insulin Delivery Systems, Pancreas and Islet Transplantation) Study Group. (Now known as AIDPIT).

The presented results of this innovative islet purification method — developed over nearly 10 years (from 1989) in our Leiden islet laboratory (LUMC) — were the outcome of work I had done spring 1998 with the human islet transplantation group of Camillo Ricordi at the Diabetes Research Institute in Miami , Florida, USA.
